Abstract: Protein-protein interaction(PPI) plays a critical role in most cellular processes and serves as a scaffold in both structural &functional organization of living system. First, protein interfaces of known 3D structure are classified according to their binding orientations. Nearly 6000 distinct interface types are classified, revealing several interesting aspects of interface evolution including interaction diversity, specificity and domain-fusion. The resulting classification (SCOPPI: Structural Classification of Protein-Protein Interfaces) has been useful as a framework for various interface studies. Second, the global structure of PPI network shows interesting characteristics such as scale-freeness (or power-law degree distribution) and hierarchical modularity. The structures of such networks underlie important features of biological systems, such as functional modularity, error-tolerance, and stability. It is not yet known how these network architectures originate or what driving forces underlie the observed network structure. Currently preferred model - gene duplication &divergence or preferential attachment model - is based only on network topologies but is not compatible with the unique age-distribution patterns observed in real yeast PPI networks.
