Regulation of Transcription elongation by chromatin remodeler

The protein complexes that control RNA polymerase II (RNAPII) elongation rate and processivity are
poorly understood. Moreover, it is unclear how chromatin remodelers affect transcription elongation
through communicating with RNAPII. To address these questions, we utilized the simple yeast genetics,
high-throughput genome analysis, and bioinformatic technique to analyze RNAPII transcription
elongation in vivo. Especially, we characterized INO80 complex and its binding partners to understand
transcription elongation mechanism. We found that INO80 regulates promoter-proximal pausing-like
pattern of RNAPII and this activity is conserved in mouse embryonic stem cells, suggesting that INO80
complex orchestrates a mechanism for proper transcription elongation, linking chromatin remodeling
activity to productive mRNA synthesis mediated by RNAPII. In this talk, I will discuss the recent
progress of our laboratory about chromatin remodeling complex that function within a defined window to
promote Pol II processivity, elongation rate variation, and progression past polyadenylation sites in early
gene body regions to prevent accumulation of immature, truncated transcripts.
